hi rob- in the middle of harvesting and reusing yeast myself. found mr malty a little difficult to understand and didnt really give me what I was looking for- http://yeastcalc.com/ tells you how much you'll need and how big a starter will give you it

also, for those test tubes, wyeast have some really interesting info- goto the bottom of this page and it gives you estimates of cell counts http://www.wyeastlab.com/hb_pitchrates.cfm

You will definitely need to make a starter. Or if you are going to breww again a few weeks after the last one keep the whole yeast cake, wash it and pitch the whole lot it will usually last a few weeks in the fridge if you are meticulously about sterilization.

I'm going to have a go at freezing yeast, been reading up about it.
