So I just cropped a vial of yeast from the top of my latest brew and now have 30ml of foamy yeast in a small container, I want to grow this yeast and then split the remaining yeast into 4 vials for storage and later use...here's my proposed method...your comments/advice will be greatly received :)

Step 1:

Add the 30ml yeast vial into 100ml of 1020 LDME wort - leave for 24 hours @ 18c

Step 2:

Add this to 250ml of 1020 LDME wort - leave for 24/48 hours @ 18c

Step 3:

Add this to 1 litre of 1020 LDME wort - leave for 24/48 hours @ 18c then place in the fridge @ 1-2c to allow yeast to drop out of liquid.

Step 4:

drain off the liquid and then split the remaining slurry into 4 sterilised vials for later use (which I will make a starter from before pitching, using steps 1-3 above)

Why don't you just crop more yesst. I usually get about a litre of settled yesst of a brew.

Two reasons...I kind of want to give this a go in order to get used to handling the yeast as I've never top cropped before and also want to try out growing the yeast from a small amount...its more as a learning experience really :)

I'd use the x10 rule and also use a wort more like 1040 (liquor to DME =10:1 = 1L to 100g DME) also ferment between 18-24C, so by all means start with 30ml yeast slurry with 300ml wort, then decant spent wort and step up with 3L of wort the decant and split 4 ways. The less processes you do the less chance of infection.

As grays said it would be easier to just top crop 4 vials.

Also there is no substitute for the Yeast book.

Good luck

I thought the yeast book says to use a lower SG wort so as not to stress the yeast you don't want it to produce alcohol just reproduce and metabolise aerobically. :thumb:

no they recommend 1030 to 1040 for the starter wort using a 10:1 ratio wort volume:DME, you would only go down to 1020 if you were culturing from a bottle conditioned beer or an old yeast
