Is is possible to use only some of the vial of a liquid yest like wlp300 and reseal - making several starters as and when you need to from one vial?

I tend to make a larger starter (usually 1l larger) than I need and put the unused portion back into the vial after draining off almost all the spent wort.
 
I usually make up a 300ml starter and then step this up again to 1 litre.
Once fermented out I put it in the fridge to allow it to settle for 2 days, decant the spent wort and then top up with distilled water that has been boiled and cooled.
Re-suspend the yeast in this and then decant in to 4 bottles - usually 330ml glass bottles that I can crown cap.

Each brew day, I then make a starter from one of the bottles a few days in advance.

I did the same as markp and basically split 4 ways, using the first for a Belgian Strong Ale.

Then, because I was planning on doing a similar (but lower gravity) beer the following week, I cleaned up the trub after the primary and pitched a quarter of that in the new brew, keeping the rest.

I've now got 3 bottles of first-generation WLP550 and 3 bottles of second-generation WLP550 in the fridge. At this rate, by the end of a year, there will be more bottles of WLP550 in the world than there are rabbits.
