I have just primes and kegged a St Peters RRA in a new kingkeg barrel. Do I need to add CO2 from my cannister now or wait until it has conditioned and I am drinking it to maintain pressure in the barrel?

The carbonation CO2 usually keeps it going until half way empty, then one canister is usually enough to dispense the rest. So you shouldn't need to add now.
 
HB cylinder or bulbs?
If you have a cylinder give it a half second blast then wait 15mins then open the cap and blast again this expelled the air helping keep the beer longer.
Bulbs give a little too much s just expel some air after some conditioning has occurred.
